    La ricerca di conglomerati bituminosi di elevate prestazioni sovente si associa allo studio delle implicazioni ambientali che la produzione, la posa in opera e l’esercizio di tali materiali comportano. Un aspetto assai importante, in tal senso, è l’opportunità che alcune modalità produttive offrono in termini di un ridotto consumo di inerti e del contestuale impiego di materiali destinati a discarica che, invece, vengono inseriti in processi di riciclaggio. È questo il caso della gomma proveniente dal riciclaggio industriale di pneumatici dismessi che, negli Stati Uniti ed in Canada, è divenuta una fonte sostanziosa per la realizzazione di conglomerati bituminosi. La presente ricerca, dopo una preliminare e sintetica descrizione del granulato di gomma e delle sue proprietà chimico-fisiche, si propone di evidenziare sperimentalmente il comportamento meccanico di conglomerati bituminosi per strati di base confezionati in laboratorio con l’aggiunta di questo componente, mediante i noti processi Dry e Wet, elaborati e combinati secondo opportuni criteri metodologici. In particolare l’indagine verte su prove inserite nell’attuale normativa italiana, con l’obiettivo di pervenire ad un quadro rappresentativo dell’influenza del granulato sulle caratteristiche meccaniche analizzate

    Whole-genome doubling affects pre-mirna expression in plants

    Whole-genome doubling (polyploidy) is common in angiosperms. Several studies have indicated that it is often associated with molecular, physiological, and phenotypic changes. Mounting evidence has pointed out that micro-RNAs (miRNAs) may have an important role in whole-genome doubling. However, an integrative approach that compares miRNA expression in polyploids is still lacking. Here, a re-analysis of already published RNAseq datasets was performed to identify microRNAs’ precursors (pre-miRNAs) in diploids (2x) and tetraploids (4x) of five species (Arabidopsis thaliana L., Morus alba L., Brassica rapa L., Isatis indigotica Fort., and Solanum commersonii Dun). We found 3568 pre-miRNAs, three of which (pre-miR414, pre-miR5538, and pre-miR5141) were abundant in all 2x, and were absent/low in their 4x counterparts. They are predicted to target more than one mRNA transcript, many belonging to transcription factors (TFs), DNA repair mechanisms, and related to stress. Sixteen pre-miRNAs were found in common in all 2x and 4x. Among them, pre-miRNA482, pre-miRNA2916, and pre-miRNA167 changed their expression after polyploidization, being induced or repressed in 4x plants. Based on our results, a common ploidy-dependent response was triggered in all species under investigation, which involves DNA repair, ATP-synthesis, terpenoid biosynthesis, and several stress-responsive transcripts. In addition, an ad hoc pre-miRNA expression analysis carried out solely on 2x vs. 4x samples of S. commersonii indicated that ploidy-dependent pre-miRNAs seem to actively regulate the nucleotide metabolism, probably to cope with the increased requirement for DNA building blocks caused by the augmented DNA content. Overall, the results outline the critical role of microRNA-mediated responses following autopolyploidization in plants

    Demineralizing action of EGTA in endodontics.

    The demineralization of dentin obtained by treatment with a chelating agent ethylene diamminotetracetic acid (EDTA) or ethylene glycol-bis(β-aminoethyl ether)-N-tetraacetic acid (EGTA), is a dynamic process involving chelation and solubilization. The actions of the EDTA and EGTA on dentin are influenced by the pH. Increasing mM concentrations of EDTA or EGTA the equivalent pH decreases in a similar slope to 80 mM chelator concentration. Increasing the chelator concentration different data were obtained: with EGTA the pH decreases slightly while with EDTA goes back up to the initial values. After 80 mM, EDTA reduces the activity on the dentin, and EGTA continues to work at higher concentrations. We demonstrated that EGTA solubilized more of 60% of dentin while EDTA gives about 20% at the maximal of the solubility

    Genome-wide SNP discovery and population structure analysis in pepper (Capsicum annuum) using genotyping by sequencing

    BACKGROUND: Knowledge on population structure and genetic diversity in vegetable crops is essential for association mapping studies and genomic selection. Genotyping by sequencing (GBS) represents an innovative method for large scale SNP detection and genotyping of genetic resources. Herein we used the GBS approach for the genome-wide identification of SNPs in a collection of Capsicum spp. accessions and for the assessment of the level of genetic diversity in a subset of 222 cultivated pepper (Capsicum annum) genotypes. RESULTS: GBS analysis generated a total of 7,568,894 master tags, of which 43.4% uniquely aligned to the reference genome CM334. A total of 108,591 SNP markers were identified, of which 105,184 were in C. annuum accessions. In order to explore the genetic diversity of C. annuum and to select a minimal core set representing most of the total genetic variation with minimum redundancy, a subset of 222 C. annuum accessions were analysed using 32,950 high quality SNPs. Based on Bayesian and Hierarchical clustering it was possible to divide the collection into three clusters. Cluster I had the majority of varieties and landraces mainly from Southern and Northern Italy, and from Eastern Europe, whereas clusters II and III comprised accessions of different geographical origins. Considering the genome-wide genetic variation among the accessions included in cluster I, a second round of Bayesian (K = 3) and Hierarchical (K = 2) clustering was performed. These analysis showed that genotypes were grouped not only based on geographical origin, but also on fruit-related features. CONCLUSIONS: GBS data has proven useful to assess the genetic diversity in a collection of C. annuum accessions. The high number of SNP markers, uniformly distributed on the 12 chromosomes, allowed the accessions to be distinguished according to geographical origin and fruit-related features. SNP markers and information on population structure developed in this study will undoubtedly support genome-wide association mapping studies and marker-assisted selection programs.     HBV vaccination with Fendrix is effective and safe in pre-dialysis CKD population

    Background: Patients with chronic kidney disease have a poor response to hepatitis B vaccine due to the immunodeficiency conferred from chronic uremia. A recombinant HB vaccine containing an improved adjuvant system AS04 (HBV-AS04) has been manufactured but scarce evidence exists on HBV-AS04 use among patients with CKD. Aim: To assess efficacy and safety of an adjuvanted recombinant vaccine (HBV-AS04) in a large cohort of CKD patients at pre-dialysis stage (with susceptibility to HBV infection). Methods: Patients were prospectively enrolled to receive four 20-mcg doses of HBV-AS04 by intramuscular route (deltoid muscle) at months 1, 2, 3, and 4. Anti-HBs surface antibody concentrations were tested at intervals of 1, 2, 3, 4, and 12 months. Multivariate analyses were performed to assess the parameters, which predicted immunologic response to HBV-AS04 vaccine. Results: One hundred and seven patients were included and 102 completed the study. At completion of vaccine schedule, the frequency of responders (anti-HBs titers 65 10 mIU/mL) was 95% (97/102) (mean anti-HBs antibody titers, 688.9 \ub1 385 mIU/mL), according to per-protocol analysis. Serum haemoglobin levels were greater in responder than non- or low-responder patients to HBV-AS04 (P = 0.04) and this was confirmed by multivariate analysis. The seroprotection rate at month 50 was 88% (30/34) with lower anti-HBs antibody titers (218.5 \ub1 269.6 mIU/mL, P = 0.001). No major side effects were observed. Conclusions: Our prospective study performed in a real-world setting showed a high immunogenicity and safety of HBV-AS04 vaccine in patients with CKD not yet on maintenance dialysis. Studies provided with longer follow-ups are under way to assess the durability of seroprotection in responders
